Hampshire's 263 was a stop-start effort, but Jake Lehmann's 68 provided the backbone of the innings. He shared useful stands with Toby Albert, who made 46, and Ben Brown, who scored 54.
Glamorgan's depleted attack remained disciplined throughout, with Dan Douthwaite taking 3-38 while Zain Ul Hassan and part-time spinner Jack Hope-Bell claimed two wickets each.
What changes did the teams make?
Hampshire recalled Eddie Jack and Manny Lumsden, both unused in the Hundred franchise tournament, while Glamorgan handed a debut to 17-year-old Harry Wallace, the son of former club captain Mark, and brought back Colin Ingram for his first tournament appearance since the 2024 final win.
Nick Gubbins chose to bat, but he and Ali Orr both fell cheaply to Douthwaite and Ul Hassan, leaving Hampshire 18-2.
How did Hampshire recover from the early wickets?
Toby Albert and Lehmann quickly lifted the scoring rate with the first of two major partnerships, adding 78 at a run a ball against a raw teenage attack.
Albert was then smartly stumped for 46 by Alex Horton after charging at part-time off-spinner Jack Hope-Bell, who also bowled Tom Prest when he attempted a reverse sweep.
Brown, having survived a drop at square leg, added 70 with Lehmann to put the home side firmly on top. But neither batter went on to a century: Lehmann was run out by Eddie Byrom and Brown was bowled by Glamorgan captain Andy Gorvin, either side of Ul Hassan yorking Indian all-rounder Ashutosh Sharma for seven.
Useful late runs came from Andrew Neal, who made 23, Jack, who scored 19, and Abbott, who also made 19, to give Hampshire a defendable total. Tom Edwards and Douthwaite, with two wickets, helped curtail the innings.
How did Abbott and Lumsden break Glamorgan?
Glamorgan reached 24-0 before Abbott took control, with his pace proving too much for Byrom (14), Ul Hassan (11) and Wallace (3) in the space of seven deliveries as the visitors collapsed to 30-3.
Hope-Bell made 21 and started positively, but he then slashed Lumsden to point, leaving all the responsibility on Ingram. Ingram had already gone off the field earlier with a hand injury and began cautiously, aware of the burden on him.
Jack's second spell deepened Glamorgan's problems as Ingram top-edged him to square leg for 24 and Horton was trapped lbw second ball, strengthening Hampshire's grip on the game.
Callum Nicholls and Douthwaite then produced Glamorgan's only half-century stand, before Douthwaite was caught in the deep off Ashutosh Sharma for 28.
Nicholls was bowled cutting at Lumsden to end any realistic hope of a comeback as Hampshire supporters relaxed in the evening sunshine, and the 17-year-old then had Gorvin caught for 20 to finish with personal best bowling figures.
